Antibody Amplification System

Antibody amplification system is a detection method that increases the measurable signal generated by antigen-antibody binding, enabling researchers to identify low-abundance targets in immunological and infectious-disease samples. It commonly works by layering signal-generating components: a primary antibody binds the target, and labeled secondary antibodies or affinity-based reagents bind the primary antibody, while attached enzymes or fluorophores produce amplified signals. In immunology and infection research, these systems support antigen detection in cells, tissues, and diagnostic specimens, improving visualization when direct labeling yields a weak readout.

Scalable High Throughput Selection From Phage-displayed Synthetic Antibody Libraries

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.

Visualization of Mitochondrial DNA Replication in Individual Cells by EdU Signal Amplification

We developed a sensitive technique to label newly synthesized mitochondrial DNA (mtDNA) in individual cells in order to study mtDNA biogenesis. The technique combines the incorporation of EdU together with a tyramide signal amplification (TSA) protocol to visualize mtDNA replication within subcellular compartments of neurons.
