Immunosuppressed Mice

Immunosuppressed mice are laboratory models with deliberately weakened or absent immune function, enabling researchers to study host defense, infection, transplantation, and immune-dependent disease. Immunosuppression can result from genetic defects, targeted depletion of immune-cell populations, irradiation, or drugs that inhibit lymphocyte activation and proliferation, reducing rejection of transplanted cells or tissues. In immunology and infection research, these mice support the growth of human cells, tumors, or pathogens that normal mice may eliminate, allowing analysis of microbial virulence, immune evasion, therapeutic efficacy, and tissue responses. Their defined immune deficiencies help isolate specific host-pathogen interactions while highlighting the functions of intact immune pathways.

The mouse heterotopic heart transplantation model has been proven by many investigators to be an important method for studying mechanisms of rejection and immune response. However, the techniques involved are still challenging. By modifying standard techniques we have had success with more than 1000 transplants.

The mouse small bowel transplantation model has been recognized as an important tool to study mechanismes of immune rejection and screen new immunosuppressive drugs. However, this model is limited to use because the techniques involved is an extremely technically challenge. Now we introduce the modified technique.

Methods to Evaluate Cytotoxicity and Immunosuppression of Combustible Tobacco Product Preparations

Using optimized human peripheral blood mononuclear cell (PBMC) ex vivo assays, we showed that a combustible tobacco product preparation markedly suppresses receptor-mediated intracellularly secreted cytokines and cytolytic ability of effector PBMCs. These rapid assays may be useful in product evaluation and understanding the potential long-term effects of tobacco exposure.
