In Vitro Differentiation

In vitro differentiation is the controlled conversion of immature or progenitor cells into specialized cell types outside the body, enabling researchers to study cell development under defined laboratory conditions. The process uses culture environments containing selected growth factors, cytokines, extracellular-matrix components, or signaling molecules that activate lineage-specific pathways and guide changes in cell phenotype and function. In immunology and infection research, in vitro differentiation can generate immune-cell populations for examining maturation, pathogen recognition, inflammatory responses, and host–microbe interactions. These models support mechanistic studies, disease modeling, therapeutic screening, and the evaluation of how environmental cues shape immune-cell behavior.

In Vitro Culture and Differentiation of Primary Monocytes into Macrophages

2025

This video demonstrates the priming of primary monocytes into macrophages. Primary human monocytes can be isolated and differentiated to an M1-like pro-inflammatory phenotype using the granulocyte-macrophage colony-stimulating factor (GM-CSF) or to an M2-like anti-inflammatory phenotype using the macrophage colony-stimulating factor (M-CSF).

An In Vitro Method for the Differentiation of Megakaryocytes and Platelet Formation

2025

This video demonstrates a technique for the differentiation of megakaryocytes and the formation of platelets. Hematopoietic stem progenitor cells — isolated from human umbilical cord blood — differentiate into mature megakaryocytes in the presence of thrombopoietin. The mature megakaryocytes produce cytoplasmic extensions termed proplatelets, which give rise to individual platelets.

In Vitro Mouse Bone Marrow Stromal Cell Differentiation into Adipocytes

2025

This video demonstrates the differentiation of bone marrow stromal cells (BMSCs) into adipocytes. In the two-step process, in the presence of specific growth factors and hormones, undifferentiated BMSCs first differentiate into preadipocytes. The second step involving treatment with glucose and insulin leads to the formation of mature adipocytes with multiple lipid droplets.

Here, we present a protocol to efficiently generate human trophoblastic cells from human pluripotent stem cells using bone morphogenic protein 4 and inhibitors of the Activin/Nodal pathways. This method is suitable for the efficient differentiation of human pluripotent stem cells and can generate large quantities of cells for genetic manipulation.
