Kidney Mononuclear Cells

Kidney mononuclear cells are immune cells with a single, nonsegmented nucleus that reside in or can be isolated from renal tissue, where they help regulate local defense and inflammation. This heterogeneous population includes lymphocytes, monocytes, macrophages, and dendritic cells, which detect antigens or danger signals through antigen receptors and innate immune sensors, then coordinate responses through cytokine release and cell-to-cell signaling. In immunology and infection research, analyzing these cells helps reveal how pathogens, tissue injury, and inflammatory diseases affect the kidney. Their phenotypes and functions can also inform studies of renal immunity, host-pathogen interactions, and potential therapeutic targets.

DNabT cells are rare among peripheral T cells; however, they are abundant in certain non-lymphoid tissues. Difficulty of isolating DN T cells from non-lymphoid tissue hinders their functional analysis despite increasing recognized pathophysiologic significance. We describe a novel protocol for isolation of highly purified DN T cells from murine kidney.

Ex Vivo Expansion of Natural Killer Cells from Peripheral Blood Mononuclear Cells

This video demonstrates a technique for the expansion of natural killer (NK) cells directly from peripheral blood mononuclear cells (PBMCs) using a feeder cell line. PBMCs are co-cultured with genetically modified lymphoblastoid feeder cells expressing membrane-bound interleukin-21 (mIL-21), and the culture is supplemented with the cytokines interleukin-2 (IL-2) and interleukin-15 (IL-15). Together, this cocktail of ligands leads to the prolonged expansion of NK cells with high purity.

In this video, we describe a protocol for human primary T cell activation. The incubation of naïve T cells with magnetic beads coated with anti-CD3 and anti-CD28 antibodies, followed by interleukin-2 or interleukin-15 treatment, leads to the activation of naïve T cells and their differentiation and proliferation into effector or memory T cells.

In this video, we demonstrate the isolation of lamina propria mononuclear cells from murine colonic tissue by enzymatic digestion using collagenase E and silica-based density separation media gradient separation. The isolated mononuclear cells can be used for subsequent downstream analysis.
