Regulatory Myeloid Cells

Regulatory myeloid cells are innate immune cells that restrain or reshape immune responses, helping balance antimicrobial defense with tissue protection and immune tolerance. This functional group includes context-dependent macrophages, dendritic cells, and myeloid-derived suppressor cells that influence T-cell and other leukocyte activity through altered antigen presentation and mediators such as IL-10, TGF-β, arginase-1, nitric oxide, and reactive oxygen species. During infection, they can limit immunopathology and support tissue repair, but prolonged suppression may permit pathogen persistence and weaken protective immunity. Defining their signals, phenotypes, and tissue-specific functions supports research on host-pathogen interactions, inflammatory disease, vaccines, and immune-modulating therapies.

Given the limited xenograft models available to study interactions between human CART and myeloid cells, we established in vitro and in vivo models to understand the impacts of human macrophages on CART cells. Findings can potentially be generalized to evaluate macrophage roles in the tumor microenvironment and test macrophage-targeted immunotherapies.

This video demonstrates the procedure for differentiating myeloid dendritic cells from mouse bone marrow. Isolation of mouse tibia and femur, and processing of bone marrow are demonstrated. Pictures demonstrating cell morphology before and after differentiation, and figures depicting cell phenotype and IL-12 production following maturation using CpG are shown.

We describe a method for generating regulatory, memory and naïve T cells from a single human blood donor. Polarized Tregs can be then compared to other subsets in a variety of genetic and functional applications with genetic homogeneity, including a suppression assay also detailed here.

This protocol details a method to isolate antigen presenting cells from human thymus via different steps of enzymatic digestion of the tissue followed by density centrifugation of the single cell suspension and finally magnetic and/or FACS sorting of the cell populations of interest.
