T Lymphocyte Adhesion

T Lymphocyte Adhesion is the regulated attachment of T cells to vascular endothelium, antigen-presenting cells, and other tissues, a process essential for immune surveillance and defense against infection. It begins when selectins and their ligands mediate transient rolling, followed by chemokine-triggered activation of integrins such as LFA-1, which bind ICAM-1 to produce firm adhesion and support migration or stable cell-cell contact. These interactions guide T lymphocytes into infected tissues and help form immunological synapses for antigen recognition and signaling. Understanding T lymphocyte adhesion supports research on inflammation, host-pathogen interactions, immune deficiencies, and therapies that modify leukocyte trafficking.

Static adhesion assay is a powerful tool that can be used to model the interactions between T lymphocytes and other cell types. Interactions are generated by injecting labeled T cells into wells coated with adhesion molecules, while a plate reader is used to quantify the number of adherent cells following serial washes.

Assay of Adhesion Under Shear Stress for the Study of T Lymphocyte-Adhesion Molecule Interactions

This flow adhesion assay provides a simple, high impact model of T cell-epithelial cell interactions. A syringe pump is used to generate shear stress, and confocal microscopy captures images for quantification. The goal of these studies is to effectively quantify T cell adhesion using flow conditions.

With effector functions distinct from other T cell subsets, Th17 cells have been centrally implicated in inflammatory autoimmunity. This in vitro Th17 differentiation protocol provides a means to determine whether naïve CD4+ T lymphocytes can differentiate into Th17 cells, and to further examine their role in autoimmunity and host response.

Abdominal adhesions that form after surgery are a major cause of pain, infertility, and hospitalization and reoperation for small bowel obstruction. Our surgical procedure for creating abdominal adhesions in mice is a reliable tool to study the mechanisms underlying the formation of adhesions.
