Epidermal Melanin Induction

Epidermal melanin induction is the process by which skin increases melanin production to influence pigmentation and help protect epidermal cells from ultraviolet radiation. Ultraviolet exposure can trigger keratinocyte signaling, including p53-dependent production of melanocortins such as α-melanocyte-stimulating hormone, which activates MC1R on melanocytes and increases tyrosinase activity; the resulting melanin is packaged into melanosomes and transferred to keratinocytes. In medicine, studying this pathway supports research on tanning responses, photoprotection, hyperpigmentation, and pigmentary disorders. It also provides a framework for evaluating agents that regulate melanogenesis and for developing safer approaches to managing abnormal skin pigmentation.

Epidermal melanin is induced by topical application of forskolin in a murine model of the fair-skinned UV-sensitive human. Pharmacologic manipulation of cAMP levels in the skin and epidermal darkening strongly protect against UV-mediated inflammation (sunburn) as measured by the minimum erythematous dose (MED) assay.

Here we show our method to isolate mouse epidermal neural crest stem cells (EPI-NCSC). Technique involves micro-dissecting whisker follicles, isolating the bulge and placeing it into tissue culture. EPI-NCSC start to emigrate from bulge explants onto the substratum within 3 - 4 days.
