Blood Chimerism

Blood chimerism is the coexistence of genetically distinct blood-cell populations within one person, a condition that can result from hematopoietic stem-cell transplantation, pregnancy, or other forms of cell exchange. In clinical practice, laboratories assess the proportion of donor- and recipient-derived cells by analyzing genetic markers, often short tandem repeats, in blood samples over time. This measurement helps determine whether transplanted stem cells have successfully established donor hematopoiesis, identify mixed or declining engraftment, and support evaluation of relapse or graft failure. Blood chimerism analysis therefore guides post-transplant monitoring and informs treatment decisions in transplantation medicine.

We describe a protocol whereby busulfan conditioning permits the bone marrow of a recipient mouse to be replaced with bone marrow cells from donor mice ubiquitously expressing green fluorescent protein, in the absence of irradiation. This technique is useful to study bone marrow cell accumulation in the central nervous system.
