Evans Blue Standard Curve

An Evans Blue standard curve is a calibration graph that relates known dye concentrations to measured optical signals, enabling quantitative analysis in biomedical samples. It is generated by preparing serial dilutions of Evans Blue, measuring their absorbance with a spectrophotometer, and plotting signal against concentration to establish a working relationship for calculating unknown values. Because Evans Blue binds strongly to serum albumin and can serve as a tracer of vascular permeability, the curve supports estimation of dye or albumin leakage in plasma, tissues, and experimental models. In medicine, this quantitative approach helps assess barrier integrity, edema, inflammation, and treatment-related changes in microvascular function.

In this study, we describe a straightforward method to perform Evans Blue Dye (EBD) analysis on zebrafish larvae. This technique is a powerful tool for the characterization of skeletal muscle integrity and delineation of zebrafish models of muscular dystrophy, and is a valuable method for the development of novel therapeutics.
