Fresh Tissue Analysis

Fresh tissue analysis is the examination of newly collected biological tissue before fixation or extensive processing, helping preserve features that may be altered during conventional preparation. Depending on the clinical question, specimens can undergo gross assessment, rapid sectioning, microscopy, or molecular testing while their native cellular and biochemical characteristics remain available for evaluation. In medicine, this approach supports diagnostic assessment, biomarker analysis, surgical decision-making, and investigation of disease mechanisms. By linking tissue structure with molecular findings, fresh tissue analysis can provide timely information for patient care and generate research material for studying cancer, infection, inflammation, and other pathological conditions.

A Simple and Rapid Protocol to Non-enzymatically Dissociate Fresh Human Tissues for the Analysis of Infiltrating Lymphocytes

This protocol describes the rapid non-enzymatic dissociation of fresh human tissue fragments for qualitative and quantitative assessment of CD45+ cells (lymphocytes/leukocytes) present in various normal and malignant human tissues. Additionally, the supernatant obtained from the primary tissue homogenate can be collected and stored for further analysis or experimentation.

Here we present a Fluorescence Activated Cell Sorting (FACS) protocol to study molecular alterations in Fos-expressing neuronal ensembles from both fresh and frozen brain tissue. The use of frozen tissue allows FACS isolation of many brain areas over multiple sessions to maximize the use of valuable animal subjects.
