Ige-mediated Allergy

IgE-mediated allergy is an immune response in which otherwise harmless substances, such as foods, drugs, or environmental allergens, trigger immediate hypersensitivity and potentially severe symptoms. During sensitization, allergen-specific IgE antibodies bind high-affinity FcεRI receptors on mast cells and basophils; subsequent allergen exposure cross-links these antibodies, causing rapid degranulation and release of histamine, leukotrienes, and other inflammatory mediators. This mechanism underlies conditions including allergic rhinitis, asthma, urticaria, and anaphylaxis, and supports diagnostic testing that detects allergen-specific IgE. Understanding the pathway also guides treatments such as allergen avoidance, antihistamines, epinephrine, and targeted biologic therapies.

Basophil activation test is a potent tool for the detection of IgE-dependent allergies in vitro. Here, an optimized protocol for basophil activation test is used to investigate drug hypersensitivity. A method for the efficient production of covalent drug-protein conjugates and their physicochemical characterization is described.

In vitro analysis of class switch recombination in mice is challenging due to cytophilic IgE molecules bound to Fc receptors on the surface of B cells. We describe a method for IgE detection using trypsin-mediated cleavage of surface-bound IgE prior to fixation and permeabilization for cytoplasmic fluorescence staining.

Here we present a new method to accurately measure body temperature differences in passive systemic anaphylaxis (PSA) and food allergy mouse models using an infrared thermometer. This procedure has been accurately duplicated in previous PSA results.
