Ionized Calcium Analysis

Ionized calcium analysis measures the free, biologically active fraction of calcium in blood, providing a direct assessment of calcium availability for neuromuscular, cardiac, and cellular functions. The test typically uses an ion-selective electrode to detect calcium ions in a blood sample, while accounting for conditions such as pH that alter calcium binding to proteins. Unlike total calcium measurement, ionized calcium analysis can reveal clinically important disturbances when albumin levels or acid-base status are abnormal. It supports evaluation and management of hypocalcemia, hypercalcemia, critical illness, kidney disease, and disorders of parathyroid regulation.

Ionization Energy

The amount of energy required to remove the most loosely bound electron from a gaseous atom in its ground state is called its first ionization energy (IE1). The first ionization energy for an element, X, is the energy required to form a cation with 1+ charge: The energy required to remove the second most loosely bound electron is called the second ionization energy (IE2). The energy required to remove the third electron is the third ionization energy, and so on. Energy is always required...

Source: Hakim, M. A. et al., Isolation and Functional Analysis of Arteriolar Endothelium of Mouse Brain Parenchyma. J. Vis. Exp. (2022).This video demonstrates the functional analysis of an arteriolar endothelial tube isolated from a mouse brain. It outlines the steps for measuring intracellular calcium levels and membrane potential, both at baseline and in response to pharmacological stimulation.

Changes in the intracellular calcium levels in the podocytes are one of the most important means to control the filtration function of glomeruli. Here we explain a high-throughput approach that allows detection of real-time calcium handling and single ion channels activity in the podocytes of the freshly isolated glomeruli.

Gas Chromatography (GC) with Flame-Ionization Detection

Source: Laboratory of Dr. B. Jill Venton - University of Virginia Gas chromatography (GC) is used to separate and detect small molecular weight compounds in the gas phase. The sample is either a gas or a liquid that is vaporized in the injection port. Typically, the compounds analyzed are less than 1,000 Da, because it is difficult to vaporize larger compounds. GC is popular for environmental monitoring and industrial applications because it is very reliable and can be run nearly continuously.

Calcium Imaging in Neurons

Calcium ions play an integral role in neuron function: They act as intracellular signals that can elicit responses such as altered gene expression and neurotransmitter release from synaptic vesicles. Within the cell, calcium concentration is highly dynamic due to the presence of pumps that selectively transport these ions in response to a variety of signals. Calcium imaging takes advantage of intracellular calcium flux to directly visualize calcium signaling in living neurons. This video begins...
