Leukocyte Rolling Quantification

Leukocyte Rolling Quantification is the measurement of how white blood cells transiently tether to and roll along the vascular endothelium, a key early step in inflammation and immune surveillance. Under blood-flow shear, selectins on endothelial cells bind carbohydrate ligands on leukocytes, creating rapid cycles of attachment and release that slow cells without stopping them completely. Researchers quantify rolling velocity, frequency, duration, and the proportion of interacting cells using live-cell imaging or flow-based assays. These measurements help evaluate endothelial activation, leukocyte adhesion, and inflammatory responses, supporting studies of vascular disease, infection, immune disorders, and therapies that target leukocyte recruitment.

This report provides a visual depiction of parallel-plate flow chamber analysis for studying leukocyte endothelial interactions under physiologic shear stress. This method is particularly useful for investigating the role of endothelial (E)-selectin and leukocyte E-selectin ligands that trigger leukocyte rolling on endothelial cell surfaces.

We demonstrate a method to label the walls of the retinal vasculature and adherent leukocytes. These adherent leukocytes can then be counted under a fluorescence microscope as a parameter of inflammation or the response of that inflammation to therapies.

Here, we describe a non-invasive two-photon (2P) microscopy approach to study leukocyte homing in the mouse footpad. We discuss the technical aspects of our tissue imaging preparation and walk the reader through a typical experiment from initial set up to execution and data collection.
