Lung Tissue Imaging

Lung tissue imaging comprises methods that visualize the structural and molecular features of pulmonary tissue, supporting the study and diagnosis of respiratory disease. Depending on the modality, images are formed by detecting differences in X-ray attenuation, light scattering, or fluorescence from labeled cells and tissue components, often with sectioning or three-dimensional reconstruction to resolve alveoli, airways, and blood vessels. These approaches help researchers examine inflammation, fibrosis, infection, tumors, and tissue remodeling at scales ranging from whole organs to individual cells. In medicine, lung tissue imaging links microscopic pathology with clinical findings and supports disease characterization, treatment assessment, and the development of improved diagnostic tools.

We describe a relatively simple method for ex vivo live imaging of the tumor cell-stroma interactions within lung metastasis, utilizing fluorescent reporters in mice. Using spinning-disk confocal microscopy, this technique enables visualization of live cells for at least 4 hr and could be adapted to study other inflammatory lung conditions.
