Multimodal Molecular Imaging

Multimodal molecular imaging is the integrated use of two or more imaging technologies to visualize molecular processes alongside anatomy or physiology, providing a more complete view of disease. It works by combining complementary signals, often from targeted molecular probes with modalities that offer high spatial resolution or functional information, and aligning the resulting datasets for joint interpretation. In medicine, this approach can help localize disease, characterize biological activity, guide treatment selection, and monitor response over time. By linking molecular changes with anatomical context, multimodal molecular imaging supports earlier diagnosis, more precise therapy planning, and research into disease mechanisms.

Biofunctionalized Prussian Blue Nanoparticles for Multimodal Molecular Imaging Applications

This protocol describes the synthesis of biofunctionalized Prussian blue nanoparticles and their use as multimodal, molecular imaging agents. The nanoparticles have a core-shell design where gadolinium or manganese ions within the nanoparticle core generate MRI contrast. The biofunctional shell contains fluorophores for fluorescence imaging and targeting ligands for molecular targeting.

We detail a new near-infrared fluorescence (NIRF) catheter for 2-dimensional intravascular molecular imaging of plaque biology in vivo. The NIRF catheter can visualize key biological processes such as inflammation by reporting on the presence of plaque-avid activatable and targeted NIR fluorochromes. This article describes an optimized sequence of events for multimodal imaging of cellular grafts in rodent brain using: (i) in vivo bioluminescence and magnetic resonance imaging, and (ii) post mortem histological analysis. Combining these imaging modalities on a single animal allows cellular graft evaluation with high resolution, sensitivity and specificity.
