Splenic Vessel Ligation

Splenic vessel ligation is a surgical technique that closes the splenic artery, splenic vein, or both to control blood flow to and from the spleen. During the procedure, the surgeon identifies and isolates the target vessel, then secures it with sutures, clips, or another ligature before division or continued tissue handling; interrupting arterial inflow limits bleeding, while venous ligation prevents blood drainage and may cause temporary congestion. The technique supports splenectomy, management of traumatic or operative hemorrhage, and selected pancreatic or vascular procedures. Accurate knowledge of splenic vascular anatomy is essential for effective hemostasis and for limiting unintended ischemia or injury to nearby organs.

DNA Ligation Reactions

In molecular biology, ligation refers to the joining of two DNA fragments through the formation of a phosphodiester bond. An enzyme known as a ligase catalyzes the ligation reaction. In the cell, ligases repair single and double strand breaks that occur during DNA replication. In the laboratory, DNA ligase is used during molecular cloning to join DNA fragments of inserts with vectors – carrier DNA molecules that will replicate target fragments in host organisms. A procedure for making en face preparations of the mouse carotid artery and aorta is described. Such preparations, when immunofluorescently stained with specific antibodies, enable us to study localization of proteins and identification of cell types within the entire vascular wall by confocal microscopy.

Surgical Injury to the Mouse Pancreas through Ligation of the Pancreatic Duct as a Model for Endocrine and Exocrine Reprogramming and Proliferation

This protocol describes an injury model involving the surgical ligation of the pancreatic duct in the adult mouse pancreas, resulting in severe injury that establishes an environment that allows beta cell neogenesis and proliferation. This model can be used as a tool to study mechanisms involved in beta cell formation.

The surgical procedure used to induce experimental myocardial infarction in mice begins with left thoracotomy between the third and the fourth ribs in order to visualize the anterior surface of the heart and left lung. The left coronary artery is ligated, the chest is closed and the mouse is allowed to recover spontaneously.
