Up-down Test

The up-down test is an adaptive method used in medicine and pharmacology to estimate a dose associated with a defined response, often the median effective or toxic dose, while limiting the number of subjects tested. Participants or experimental animals receive doses sequentially, and the next dose is increased after a negative response or decreased after a positive response according to a predetermined interval. This staircase design concentrates observations near the response threshold rather than testing many fixed dose groups. Up-down testing supports acute toxicity assessment, dose-finding, and evaluation of drug potency, providing a practical balance between statistical estimation, experimental efficiency, and ethical reduction of animal use.

Here, we present a protocol to use a curvelet transform-based, open-source MATLAB software tool for quantifying fibrillar collagen organization in the extracellular matrix of both normal and diseased tissues.
