Awake Animal Injection

Awake animal injection is a research technique for administering drugs, tracers, or other experimental substances to conscious animals, particularly when anesthesia could alter neural or behavioral responses. The procedure involves selecting an appropriate injection route, preparing and restraining the animal, and delivering a controlled dose while maintaining careful monitoring and minimizing stress. In neuroscience, this approach supports studies of drug effects, neural signaling, behavior, and brain–body interactions under more physiologically representative conditions. Comparing responses in awake animals can help researchers distinguish treatment effects from changes caused by anesthetic suppression, improving interpretation of behavioral and pharmacological experiments.

Electrophysiological characterization of neuronal responses is important for understanding brain function and for guiding the placement of dyes for pathway tracing. However, many studies are performed in anesthetized animals. To understand brain function without anesthetics, we developed a method to record neuronal response properties and inject dyes in awake mouse.

Here, we show the pressure injection of neuropharmacological substances during single-cell recording in an awake, behaving macaque monkey. This procedure allows pharmacological manipulation in the direct vicinity of a cortical recording site.

Implanting organized arrays of microwires for use in single-unit electrophysiological recordings presents a number of technical challenges. Methods for performing this technique and the equipment necessary are described. Also, the beneficial use of organized microwire arrays to record from distinct neural subregions with high spatial selectivity is discussed.
