Biolistic Transfection

Biolistic transfection is a physical gene-delivery technique that introduces nucleic acids into cells, making it useful when conventional chemical or viral methods are difficult to apply. In this process, DNA or other genetic material is coated onto microscopic gold or tungsten particles and propelled into target cells, where the delivered constructs can support transient or stable gene expression. In neuroscience, researchers use biolistics to label neurons, express fluorescent reporters, manipulate neuronal proteins, and study cellular morphology or circuit connectivity in cultured tissue and, in some settings, intact brain samples. Its ability to reach localized groups of cells supports spatially targeted experiments in neural development and function.

Transgenic worms are commonly used in C. elegans research. Described is a simple, yet effective, protocol to introduce transgenes into worms using biolistic bombardment with DNA-coated gold particles. The effort involved and results of bombardment compare favorably with microinjection for the generation of transgenic animals.
