Bisulfite Treatment

Bisulfite treatment is a chemical method used to distinguish methylated from unmethylated cytosines in DNA, making it essential for studying epigenetic regulation. During the process, sodium bisulfite deaminates unmethylated cytosine, converting it to uracil, while 5-methylcytosine remains largely unchanged; subsequent PCR and sequencing reveal the original methylation pattern as sequence differences. In neuroscience, this approach helps map DNA methylation across neuronal genes and regulatory regions, supporting research on brain development, synaptic plasticity, aging, and neurological disease. The resulting methylation profiles can clarify how environmental signals and cellular states influence gene expression in the nervous system.

Here, we present a protocol to remove aldehydes and reactive ketones from mixtures by a liquid-liquid extraction protocol directly with saturated sodium bisulfite in a miscible solvent. This combined protocol is rapid and facile to perform. The aldehyde or ketone can be re-isolated by the basification of the aqueous layer.
