Hippocampal Slice Preparation

Hippocampal slice preparation is a laboratory method for isolating thin sections of hippocampal tissue so researchers can study neural circuits under controlled conditions. During the procedure, brain tissue is rapidly removed, cooled, and cut with a vibratome in oxygenated artificial cerebrospinal fluid, helping preserve neuronal viability and local synaptic connections. Researchers maintain the slices in physiological solution and examine electrical activity, synaptic transmission, and plasticity using electrophysiology, imaging, or pharmacological experiments. This preparation provides a tractable model for investigating learning and memory, cellular mechanisms of neurological disease, and the effects of drugs on hippocampal function.

Organotypic Hippocampal Slice Cultures

We describe a method to prepare organotypic hippocampal slices that can be easily adapted to other brain regions. Brain slices are laid on porous membranes and culture media is allowed to form an interface. This method preserves the gross architecture of the hippocampus for up to 2 weeks in culture.
