Monolayer Culture

Monolayer culture is a laboratory method in which cells grow as a single, adherent layer on a suitable substrate under controlled conditions, enabling direct observation and experimental manipulation. In neuroscience, neurons, neural progenitor cells, or glial cells attach to the culture surface, receive defined nutrients and growth factors, and extend processes while remaining accessible for imaging, staining, electrophysiology, or molecular analysis. This approach supports studies of neuronal development, neurite outgrowth, synapse formation, cell signaling, and neurotoxicity. Although it may not reproduce the three-dimensional organization of brain tissue, monolayer culture offers a relatively simple, reproducible platform for investigating neural mechanisms and testing treatments.

Generating Neural Progenitors from Embryonic Stem Cells in Serum-Free Monolayer Cultures

The video demonstrates a protocol that uses serum-free media to generate neural progenitors from mouse embryonic stem cells. The desired concentration of embryonic stem cells, suspended in serum-free media, is plated on gelatin-coated coverslips. At an optimal cell density, cells produce autocrine signaling factors, which, along with the neural differentiation-inducing factors in the serum-free media, trigger the differentiation of embryonic stem cells into neural progenitors.

This protocol describes in detail the method for generating neural progenitors from embryonic stem cells using a serum-free monolayer method.
