Tumor Cell Isolation

Tumor cell isolation is the process of separating cancer cells from a tumor or surrounding biological material so they can be studied as a defined population. It typically involves mechanically and enzymatically dissociating tissue into individual cells, followed by enrichment or selection based on properties such as cell-surface markers, size, density, or attachment behavior. In neuroscience, isolating tumor cells from brain or nervous-system tumors supports analysis of cellular heterogeneity, neural interactions, gene expression, and treatment responses. The resulting cells can be used in culture, molecular assays, drug screening, and investigations of invasion or recurrence, helping researchers connect tumor behavior with disease mechanisms and therapeutic outcomes.

This article describes the technique for isolating pancreatic cancer stem cells (CSCs) from human pancreatic ductal adenocarcinoma (PDAC) tissue. These CSCs are used to study the functional effects of drug treatment on the in vitro self-renewal capacity of CSCs for developing more efficient cancer treatments.

The goal of this protocol is to study breast tumorigenesis. With this technique, mouse mammary tumors are removed and primary cells are prepared from tumors. A lung extraction protocol is included for studying lung metastasis. Furthermore, another protocol for analyzing mouse embryonic fibroblasts from the mouse embryo is included.
