Metabolite Neutralization

Metabolite neutralization is the process by which the body reduces the biological activity or toxicity of compounds formed during drug metabolism, an important determinant of pharmacological response and safety. In the liver and other tissues, enzymes can transform reactive metabolites through oxidation, reduction, or hydrolysis, then attach polar groups such as glucuronide or sulfate to promote renal or biliary elimination; endogenous molecules may also bind or sequester harmful products. In pharmacology, understanding these pathways helps explain drug duration, adverse effects, interactions, and variability among patients. It also guides safer drug design and evaluation of metabolic liabilities during preclinical and clinical development.

We describe here the procedures for the extraction and purification of mRNA and metabolites from Drosophila heads. We are applying these techniques to better understand the cellular perturbations underlying neuronal degeneration. These methodologies can be easily scaled and adapted for other "omic" projects.

Optimization of a Quantitative Micro-neutralization Assay

This study describes an imaging-based micro-neutralization assay to analyze the antigenic relationships between viruses. The protocol employs a flatbed scanner and has four steps, including titration, titration quantitation, neutralization, and neutralization quantitation. The assay works well with current circulating influenza A(H1N1)pdm09, A(H3N2), and B viruses.
