Pathogen Growth

Pathogen growth is the increase in infectious microorganisms or their populations through replication, a process central to understanding disease progression and treatment. Bacteria and fungi multiply when nutrients, temperature, pH, oxygen, and moisture support cellular metabolism, whereas viruses replicate by entering host cells and directing host machinery to produce new viral particles. In pharmacology, measuring growth under controlled conditions helps researchers evaluate antimicrobial activity, determine susceptibility and minimum inhibitory concentrations, and compare treatment strategies. These analyses guide drug discovery, dosing decisions, resistance monitoring, and the development of therapies that suppress pathogens while limiting harm to host tissues.

We describe a method for tracking the endomembrane rupture elicited by the intracellular bacteria Shigella flexneri and Mycobacterium tuberculosis upon host cell invasion. Our assay makes use of CCF4, a host cytoplasmic FRET probe in live or fixed cells. This reporter is degraded by an enzyme activity present on the bacterial surface.

A PCR-based protocol was adapted to detect Cronobacter spp., Salmonella enterica, and Listeria monocytogenes from body surfaces and alimentary canals of individual wild-caught flies. The goal of this protocol is to detect and isolate bacterial pathogens from individual insects collected as part of an environmental sampling program during foodborne outbreak investigations.
