Platelet-derived Growth Factor

Platelet-derived Growth Factor (PDGF) is a family of dimeric signaling proteins that regulates cell growth, survival, migration, and tissue repair, making it an important target in pharmacology. Secreted by platelets and other cells, PDGF binds PDGF receptors, which are receptor tyrosine kinases; receptor dimerization and autophosphorylation activate pathways controlling proliferation, chemotaxis, and extracellular matrix production. Pharmacological studies examine PDGF signaling in wound healing, vascular development, fibrosis, and cancer, where excessive activity can promote pathological cell growth and tissue remodeling. Drugs that inhibit PDGF receptors or downstream signaling may therefore help manage diseases driven by abnormal PDGF activity.

This article outlines the protocol for quantifying the percentage of Tissue Factor (TF)-positive platelets using whole blood flow cytometry, assessing the protein: (1) intracellularly in resting conditions, and (2) on the cell surface, in both resting and activated conditions. Guidance is also provided for evaluating TF-positive platelets in platelet-rich plasma.

We describe a simple cell-based bioassay for detecting, quantifying and monitoring the activity of members of the vascular endothelial growth factor family of ligands. The assay uses chimeric receptors expressed in a factor-dependent cell line to provide a semi-quantitative or quantitative assessment of receptor binding and cross-linking by the ligand.

Turbidimetry on Human Washed Platelets: The Effect of the Pannexin1-inhibitor Brilliant Blue FCF on Collagen-induced Aggregation

We describe a straightforward method for the isolation of washed platelets from human blood followed by agonist-induced platelet aggregation measurements by turbidimetry. As an example we apply this method for studying the aggregation response of human platelets to collagen after a pre-incubation with the Pannexin1 channel inhibitor Brilliant Blue FCF.
