Intestinal Barrier

The intestinal barrier is a selectively permeable interface that separates the gut lumen from underlying tissues while regulating nutrient absorption and limiting exposure to harmful substances. It relies on a mucus layer, intestinal epithelial cells, and tight junctions that control paracellular movement, alongside immune defenses that detect and respond to luminal signals. In pharmacology, barrier properties influence how orally administered drugs are absorbed, distributed across intestinal tissues, and affected by transporters or epithelial injury. Studying barrier permeability and its disruption helps evaluate drug safety, understand inflammation-related changes in drug response, and guide the development of targeted oral therapies.

Current treatments of inflammatory bowel diseases (IBD) aim at alleviating disease symptoms but may cause severe side effects. Thus, alternative strategies are being investigated in animal colitis models. Here, we explain how the beneficial effects of diet supplements on clinical IBD signs are analyzed in such a colitis model.

Investigating Intestinal Barrier Breakdown in Living Organoids

Here we describe a technique to quantify the barrier integrity of small intestinal organoids. The fact that the method is based on living organoids enables the sequential investigation of different barrier integrity modulating substances or combinations thereof in a time-resolved manner.

Catheterization of Intestinal Loops in Ruminants

We describe a novel surgical method for catheterizing 'intestinal loops' within the ileum of sheep. Once animals have recovered from surgery and have cleared antibiotics and analgesics, multiple treatments can be deposited directly in loops via the catheters.
