Palmitic Acid Assay

Palmitic acid assay is an analytical method used to detect and quantify palmitic acid, a saturated 16-carbon fatty acid involved in lipid metabolism and cellular energy storage. Depending on the assay design, the sample is prepared to release or extract fatty acids, and palmitic acid is measured through a selective chemical, enzymatic, or instrumental signal compared with standards of known concentration. These assays support studies of fatty acid synthesis, oxidation, membrane composition, and metabolic regulation. Reliable quantification also helps evaluate changes in lipid profiles in cells, tissues, biological fluids, and biochemical experiments.

We describe here a multiplex caspase-3/7 activity assay, using fluorescent- and luminescent-based methods, to determine cell viability in an in vitro hypothalamic model following oxidative challenge with palmitic acid.

De novo lipogenesis and β-fatty acid oxidation constitute key metabolic pathways in hepatocyte, pathways that are perturbed in several metabolic disorders, including fatty liver disease. Here we demonstrate isolation of mouse primary hepatocytes and describe quantification of β-fatty acid oxidation and lipogenesis.
