Seed Metabolite Mapping

Seed metabolite mapping is the analysis and visualization of small molecules across distinct regions of a seed, linking chemical composition to tissue structure and function. In biochemistry, researchers collect spatially resolved measurements, often by mass spectrometry imaging or targeted sampling, and assign metabolite identities from mass-to-charge signals, standards, or complementary analytical data; the resulting maps show how compounds are distributed among tissues. This approach can reveal localized reserves, signaling molecules, defense compounds, and metabolic changes during maturation or germination, supporting studies of seed quality, plant development, stress responses, and the nutritional or agricultural value of crops.

Begin with a bacterial biosensor culture suspended in a medium. These engineered bacteria produce a transcription factor that, upon binding to a specific fecal metabolite, activates a promoter associated with a fluorescent reporter gene.To this culture, add a filtered human fecal sample containing metabolites, such as lactate, that are involved in host-microbiome interactions.Dilute this mixture with a fresh medium to maintain an optimal reaction volume.Incubate without shaking to mimic the...

We describe here the procedures for the extraction and purification of mRNA and metabolites from Drosophila heads. We are applying these techniques to better understand the cellular perturbations underlying neuronal degeneration. These methodologies can be easily scaled and adapted for other "omic" projects.

Seed Structure and Early Development of the Sporophyte

2020

Seed structures are composed of a protective seed coat surrounding a plant embryo, and a food store for the developing embryo. The embryo contains the precursor tissues for leaves, stem, and roots. The endosperm and cotyledons—seed leaves—act as the food reserves for the growing embryo. The embryo contains a double set of chromosomes, one set from each parent. Fertilization of the haploid egg by the haploid sperm gives rise to the zygote, which develops into the embryo. The endosperm is a...
