High Throughput Expression

High-throughput expression is a bioengineering approach for producing and evaluating many recombinant proteins or other gene products in parallel, accelerating the search for useful biological functions. It combines parallelized cloning, host-cell transformation, and expression cultures with automated liquid handling, microplate formats, and assay-based screening. By varying genes, vectors, host strains, or induction conditions across many small-scale reactions, researchers can compare expression levels, solubility, activity, and stability efficiently. This workflow supports enzyme engineering, protein characterization, synthetic biology, and biomanufacturing by identifying promising candidates before larger-scale production, while reducing material use, manual labor, and experimental time.

We present a microfluidic approach for the expression of protein arrays. The device consists of thousands of reaction chambers controlled by micro-mechanical valves. The microfluidic device is mated to a microarray-printed gene library. These genes are then transcribed and translated on-chip, resulting in a protein array ready for experimental use.
