Methylated Collagen

Methylated collagen is collagen chemically modified with methyl groups to alter its molecular and material properties, making it relevant to bioengineering and biomaterial design. Methylation can change collagen’s polarity, charge distribution, intermolecular interactions, and susceptibility to enzymatic or chemical modification, which may influence its solubility, structure, and stability. The creation of functional microtissues within microfluidic devices requires the stabilization of cell phenotypes by adapting traditional cell culture techniques to the limited spatial dimensions in microdevices. Modification of collagen allows the layer-by-layer deposition of ultrathin collagen assemblies that can stabilize primary cells, such as hepatocytes, as microfluidic tissue models.

DNA Methylation: Bisulphite Modification and Analysis

The gold standard for DNA methylation analysis is genomic sequencing of bisulphite converted DNA. This method takes advantage of the increased sensitivity of cytosine compared with 5-methylcytosine (5-MeC) to bisulphite deamination under acidic conditions. Unmethylated cytosines can be distinguished from methylated cytosines after PCR amplification of the target genomic DNA.
