Perfusion Decellularization

Perfusion decellularization is a bioengineering technique that removes cells from an organ or tissue while preserving its three-dimensional extracellular matrix and vascular architecture. In this process, detergents, enzymes, and washing solutions are circulated through the tissue’s vascular network, allowing controlled chemical and mechanical disruption of cellular membranes and nucleic acids while maintaining structural components such as collagen and elastin. The resulting acellular scaffold can retain tissue-specific geometry and biochemical cues, making it useful for studying matrix biology and developing engineered grafts. Subsequent recellularization with appropriate cells may support efforts to create more physiologically relevant tissues for regenerative medicine and transplantation research.
