Tissue Alignment Methods

Tissue alignment methods are bioengineering strategies that organize cells, extracellular matrix, or engineered fibers along defined directions to reproduce the structural anisotropy of native tissues. These methods guide alignment through physical cues such as patterned substrates, aligned biomaterials, controlled fluid shear, mechanical strain, or electrical stimulation, which influence cell adhesion, migration, and cytoskeletal organization. Researchers use aligned architectures to improve the function of engineered muscle, nerve, tendon, ligament, and vascular tissues, where direction-dependent structure supports force transmission and signal conduction. By linking material design with cellular behavior, tissue alignment methods can enhance biomimetic models, regenerative therapies, and studies of how tissues develop and repair.

Generation of Aligned Functional Myocardial Tissue Through Microcontact Printing

The generation of aligned myocardial tissue is a key requirement for adapting the recent advances in stem cell biology to clinically useful purposes. Herein we describe a microcontact printing approach for the precise control of cell shape and function. Using highly purified populations of embryonic stem cell derived cardiac progenitors, we then generate anisotropic functional myocardial tissue.

We showcase the development of self-assembled, three-dimensional bundles of longitudinally aligned astrocytic somata and processes within a novel biomaterial encasement. These engineered "living scaffolds", exhibiting micron-scale diameter yet extending centimeters in length, may serve as test-beds to study neurodevelopmental mechanisms or facilitate neuroregeneration by directing neuronal migration and/or axonal pathfinding.

This protocol describes the isolation of dorsal root ganglion (DRG) neurons isolated from rats and the culture of DRG neurons on a static pre-stretched cell culture system to enhance axon alignment, with subsequent co-culture of Schwann Cells (SCs) to promote myelination.
