Alveolar Lung Tissue

Alveolar lung tissue is the specialized, highly vascularized region of the lungs where oxygen enters the blood and carbon dioxide is removed, making it essential for aerobic life. Its thin-walled alveoli bring air into close contact with capillaries, allowing gases to diffuse across the alveolar-capillary barrier down differences in partial pressure; pulmonary surfactant reduces surface tension and helps keep the alveoli open. Studying this tissue supports understanding of respiratory physiology and disorders such as emphysema, pulmonary edema, and fibrosis, while also informing lung imaging, drug research, and tissue-engineering strategies.

2025

This protocol presents a flow cytometry-based method to isolate viable alveolar type II epithelial cells (AECII) from neonatal, juvenile, or adult murine lungs (infected or non-infected). This approach enables downstream molecular and/or functional studies of AECII to identify their roles during lung inflammation and infection.

Here we describe the model and approach to study functions of pulmonary alveolar macrophages in cancer metastasis. To demonstrate the role of these cells in metastasis, the syngeneic (4T1) model of breast cancer in conjunction with the depletion of alveolar macrophage with clodronate liposomes was used.
