Brightfield Illumination Selection

Brightfield illumination selection is the process of choosing and adjusting transmitted-light conditions to produce clear, informative images of biological specimens. In brightfield microscopy, light passes through the sample and is modified by differences in absorption, thickness, and refractive index before reaching the objective and camera; illumination settings therefore influence contrast, brightness, and resolution. Selecting an appropriate light intensity, condenser position, aperture, and specimen preparation helps distinguish cells, tissues, and microorganisms while limiting glare and uneven background lighting. These choices support routine observation, imaging, and quantitative analysis in cell biology, histology, microbiology, and other areas of biological research.

We present a protocol for capturing the dynamics of zebrafish larval tail fin regeneration on a whole-tissue scale using brightfield-based stereomicroscopy. This technique enables capturing the regeneration dynamics with single cell resolution. This methodology can be adapted to any stereomicroscope equipped with a CCD camera and time-lapse software.

Source: Weber, N. et. al., Natural Transformation, Protein Expression, and Cryoconservation of the Filamentous Cyanobacterium Phormidium lacuna. J. Vis. Exp. (2022)This video demonstrates a focused LED-based assay for studying phototaxis in filamentous cyanobacteria. The bacteria migrate toward the light source, forming a dense accumulation at the illuminated center, which is visualized and quantified through pixel intensity analysis of the captured image.

Highly Resolved Intravital Striped-illumination Microscopy of Germinal Centers

High-resolution intravital imaging with enhanced contrast up to 120 µm depth in lymph nodes of adult mice is achieved by spatially modulating the excitation pattern of a multi-focal two-photon microscope. In 100 µm depth we measured resolutions of 487 nm (lateral) and 551 nm (axial), thus circumventing scattering and diffraction limits.
