Calcitonin Function

Calcitonin function describes how the peptide hormone calcitonin helps regulate calcium and phosphate balance, making it important for bone remodeling and mineral homeostasis. Produced mainly by parafollicular, or C, cells of the thyroid gland, calcitonin is released when blood calcium rises and lowers it by suppressing osteoclast-mediated bone resorption while promoting greater calcium loss through the kidneys. In biology and medicine, studying calcitonin function clarifies its relationship with parathyroid hormone, supports interpretation of calcium-regulating disorders, and informs the use of calcitonin in selected conditions involving elevated calcium or bone loss.

Here we present a protocol to visualize spatial correlation of calcitonin gene-related peptide (CGRP)-immunoreactive nerve fibers and blood vessels in the cranial dura mater using immunofluorescence and fluorescent histochemistry with CGRP and phalloidin, respectively. In addition, the origin of these nerve fibers was retrograde traced with a fluorescent neural tracer.
