Cellular Metabolism

Cellular metabolism is the network of chemical reactions that enables cells to obtain energy, build cellular components, maintain internal conditions, and respond to their environment. It coordinates catabolic pathways, which break down nutrients and capture energy in ATP and reduced electron carriers, with anabolic pathways, which use that energy and precursor molecules to synthesize proteins, lipids, nucleic acids, and other biomolecules; enzymes regulate each step and connect pathways through shared intermediates. In biology, studying cellular metabolism helps explain growth, exercise, development, disease, and microbial activity, while metabolic measurements and pathway analysis support research into nutrition, cancer, diabetes, and therapeutic targets.

Autofluorescence Imaging to Evaluate Cellular Metabolism

This protocol describes fluorescence imaging and analysis of the endogenous metabolic coenzymes, reduced nicotinamide adenine (phosphate) dinucleotide (NAD(P)H), and oxidized flavin adenine dinucleotide (FAD). Autofluorescence imaging of NAD(P)H and FAD provides a label-free, nondestructive method to assess cellular metabolism.

Multimodal Optical Imaging Platform for Studying Cellular Metabolism

Utilizing a multimodal platform combining label-free optical imaging modalities, we have developed a protocol for visualizing and quantifying cellular dynamics and metabolism. Through imaging via multiphoton fluorescence, second harmonic generation, and stimulated Raman scattering microscopy, we can generate a holistic overview of the cellular and molecular environment.

Overview Metabolism represents all of the chemical activity in a cell, including reactions that build molecules (anabolism) and those that break molecules down (catabolism). Anabolic reactions require energy, whereas catabolic reactions provide it. Thus, metabolism describes how cells transform energy through a variety of chemical reactions, which are often made more efficient with the help of enzymes. Metabolic Profile Analysis of Zebrafish Embryos

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.
