Chloroform Methanol Extraction

Chloroform methanol extraction is a solvent-based method for isolating lipids and other hydrophobic molecules from biological samples, enabling their analysis by biochemical and analytical techniques. A tissue or cell lysate is mixed with chloroform and methanol, which disrupt membranes and dissolve lipids; adding water or a salt solution promotes phase separation, with most lipids collecting in the lower organic layer while polar compounds remain in the upper aqueous layer. Researchers use this approach to recover phospholipids, triglycerides, sterols, and other lipid classes from cells, tissues, and microorganisms. The extracted material supports studies of membrane composition, metabolism, signaling, and lipid-associated disease.

Extraction and Analysis of Microbial Phospholipid Fatty Acids in Soils

Phospholipid fatty acids provide information about the structure of soil microbial communities. We present methods for extraction from soil samples with a single-phase chloroform mixture, fractionation of extracted lipids using solid phase extraction columns, and methanolysis to produce fatty acid methyl esters, which are analyzed by capillary gas chromatography.

This method for the first time describes reliable experimental procedures for efficient inducible methanol-free recombinant protein production in Pichia pastoris (Komagataella phaffii), employing the carbon source regulated promoters PDCand PDF in small scale experiments while cultivation parameters can be monitored online, and a constant glycerol feed is applied.

The article describes a method that increases throughput while balancing effort and accuracy for extraction of lipids from the cell membranes of microorganisms for use in characterizing both total lipids and the relative abundance of indicator lipids to determine soil microbial community structure in studies with many samples.
