Igm Antibody

IgM antibodies are a class of immunoglobulins that provide an early, important defense against newly encountered pathogens and help initiate adaptive immunity. B cells produce IgM as membrane-bound receptors and, after activation, secrete it primarily as pentamers whose multiple antigen-binding sites promote strong binding, agglutination, and activation of the classical complement pathway. Because IgM commonly appears before other antibody classes during a primary immune response, its detection can indicate recent or ongoing infection. Studying IgM supports research in immunology, vaccine responses, pathogen diagnosis, and the development of serological assays that distinguish stages of immune exposure.

Antibody Binding Specificity for Kappa (Vκ) Light Chain-containing Human (IgM) Antibodies: Polysialic Acid (PSA) Attached to NCAM as a Case Study

We present a protocol to identify protein moieties containing antigens for human and mouse IgM antibodies with specific kappa (Vκ) light chains. This protocol is not limited to IgM class antibodies but applies to all immunoglobulin isotypes that target their antigen with sufficiently high affinity during immunoprecipitations.

We describe here a method to generate customizable antigen microarrays that can be used for the simultaneous detection of serum IgG and IgM autoantibodies from humans and mice. These arrays allow for high-throughput and quantitative detection of antibodies against any antigens or epitopes of interest.

Functional Assessment of Antibody-Secreting B Cells Using ELISpot Assay

This video demonstrates the use of the ELISpot assay to assess the function of antibody-secreting B cells, ASCs. The antibodies secreted by the ASCs interact with the membrane-bound polyclonal antibody fragments, which are detected by the enzyme-conjugated IgG or IgM antibodies. The colored spot produced in each well indicates the presence of the specific antibody's isotype, which correlates with the B cells' secretory activity.
