Il-3 Withdrawal Assay

The IL-3 Withdrawal Assay is a cell-based method used to test how dependent hematopoietic cells are on interleukin-3 (IL-3) for survival and proliferation. In this assay, IL-3-dependent cells are cultured without the cytokine, causing the loss of IL-3 receptor signaling and triggering growth arrest and, often, programmed cell death through apoptotic pathways. Researchers measure outcomes such as cell viability, apoptosis, or caspase activation to assess survival signals and genetic or pharmacological influences. The assay is widely used to study cytokine signaling, hematopoietic biology, oncogenic transformation, and candidate compounds that prevent or promote apoptosis.
