Illumina Sequencing

Illumina sequencing is a high-throughput DNA sequencing technology that determines the order of nucleotides in genetic material, enabling detailed analysis of genomes, transcriptomes, and other biological samples. The method immobilizes DNA fragments on a flow cell, amplifies each fragment into a clonal cluster through bridge amplification, and then uses sequencing by synthesis to add fluorescently labeled, reversible-terminator nucleotides one at a time. Imaging after each incorporation cycle identifies the added base, while computational analysis reconstructs the sequence. Its accuracy, scalability, and relatively low cost support applications in genomics, gene expression profiling, variant detection, metagenomics, and biomedical research.

Single Read and Paired End mRNA-Seq Illumina Libraries from 10 Nanograms Total RNA

Here we describe a method for preparation of both single read and paired end Illumina mRNA-Seq sequencing libraries for gene expression analysis based on T7 linear RNA amplification. This protocol requires only 10 nanograms of starting total RNA and generates highly consistent libraries representing whole transcripts.

We describe the methodology to more accurately assess the quality of FFPE-RNA samples, prepare sequencing libraries, and analyze the data from FFPE-RNA samples.
