Pecam1 Cd31

PECAM1 (CD31) is a transmembrane cell-adhesion glycoprotein expressed mainly on endothelial cells, platelets, and subsets of leukocytes, where it supports vascular organization and immune-cell trafficking. As a member of the immunoglobulin superfamily, PECAM1 mediates homophilic interactions between neighboring cells and transmits signals through cytoplasmic immunoreceptor tyrosine-based inhibitory motifs, influencing endothelial junctions and leukocyte transmigration. In biology and biomedical research, CD31 serves as a widely used marker for endothelial cells and blood-vessel formation in tissue staining, flow cytometry, and microscopy. Studying PECAM1 helps clarify angiogenesis, vascular integrity, inflammation, and mechanisms of immune-cell movement across the endothelium.

Purification of CD31+ Endothelial Precursor Cells Using Magnetic-Activated Cell Sorting

2025

This video demonstrates a technique for purifying CD31+ endothelial progenitor cells (EPCs) from a culture using magnetic-activated cell sorting (MACS). The cell suspension is treated with a fluorophore-conjugated antibody that binds to CD31 on the EPCs. Completing the labeling of CD31+ EPCs involves adding an antibody complex, which binds to the fluorophores, and a polymer-coated magnetic nanoparticle, which binds to the antibody complex. The labeled cells are then purified using a magnetic...

The differentiation of white and beige adipocytes from adipose tissue vascular progenitors bears potential for metabolic improvement in obesity. We describe protocols for a CD34+CD31+ endothelial cell isolation from human fat and for a subsequent in vitro expansion and differentiation into white and beige adipocytes. Several downstream applications are discussed.

We present a protocol for the analysis of coronary vessels in whole embryonic murine hearts up to E15.5, using standard immunological staining methods followed by optical clearance and confocal microscopy. This technique enables visualization of blood vessels throughout the entire heart without the need for time-consuming analysis of serial sections.

Simultaneous 3D Analysis of Cardiac Damage and Immune Response in Reperfused Acute Myocardial Infarction Using Light Sheet Fluorescence Microscopy

2025

Three-dimensional (3D) reconstruction of cardiac damage in reperfused acute myocardial infarction (repAMI) enables faithful quantification and co-localization of associated disease-affecting patterns. Here, an automatable light sheet-guided imaging approach is provided for contemporaneous measurement of cardiac damage, area at risk, and immune cell response.
