Labeling Efficiency Measurement

Labeling efficiency measurement is the quantitative assessment of how effectively a biological labeling method identifies its intended molecules, cells, or structures. It typically compares the labeled fraction with the total target population by detecting marker-associated signals through fluorescence microscopy, flow cytometry, spectroscopy, or other analytical methods, while accounting for background signal and nonspecific binding. In biology, this measurement helps researchers optimize antibody labeling, nucleic acid tagging, fluorescent probes, and cell-tracking protocols. Reliable efficiency estimates improve data interpretation, support experimental quality control, and enable meaningful comparisons among labeling conditions, samples, and imaging or analytical workflows.

Standardized Method for Measuring Collection Efficiency from Wipe-sampling of Trace Explosives

Optimized sampling protocols and the development of new wipe materials can be facilitated by standardized measurements of collection efficiency from wipe-sampling. Our approach for sampling trace explosives uses an automated device to control speed, force, and distance during wipe-sampling followed by extraction of collected explosives.

The ability to measure the kinetics of vesicle release can help provide insight into some of the basics of neurotransmission. Here we used real-time imaging of vesicles labeled with the red fluorescent dye FM 4-64 to measure the rate of presynaptic vesicle release in hippocampal neuronal cultures.

This study presents an easy-to-use, complete, and simple set of methods to label and analyze glomeruli from CUBIC-cleared mouse kidneys. Data such as glomerulus number and volume can be obtained easily and reliably using fluorescein isothiocyanate (FITC)-Dextran, light sheet fluorescence microscopy (LSFM), or common confocal microscopy and software such as Imaris.
