Monolayer Cell Preparation

Monolayer cell preparation is the process of establishing a single, evenly distributed layer of cells on a suitable culture surface for controlled study in biology. It typically involves dissociating cells from tissue or an existing culture, suspending them in growth medium, and seeding them at a density that supports attachment, spreading, and population expansion without excessive overlap. Prepared monolayers provide consistent access to nutrients and gases and enable researchers to monitor cell morphology, proliferation, migration, and responses to experimental treatments. This approach supports microscopy, cytotoxicity testing, infection studies, cell signaling research, and routine maintenance of adherent cell cultures.

Studying Lymphocyte Transmigration Across a Microvascular Endothelial Cell Monolayer

This video demonstrates an experimental investigation of lymphocyte transmigration across a blood-brain barrier (BBB) endothelium model. A monolayer of human brain microvascular endothelial cells (HBMECs) is generated in vitro, and peripheral blood mononuclear cells (PBMCs) containing lymphocytes are added to the monolayer. The cells are then incubated to allow lymphocyte transmigration, and the migrated cells are quantified using flow cytometry.

Generating Neural Progenitors from Embryonic Stem Cells in Serum-Free Monolayer Cultures

The video demonstrates a protocol that uses serum-free media to generate neural progenitors from mouse embryonic stem cells. The desired concentration of embryonic stem cells, suspended in serum-free media, is plated on gelatin-coated coverslips. At an optimal cell density, cells produce autocrine signaling factors, which, along with the neural differentiation-inducing factors in the serum-free media, trigger the differentiation of embryonic stem cells into neural progenitors.
