Mouse Pancreas Dissection

Mouse pancreas dissection is the controlled anatomical separation and examination of the pancreas from a mouse, a procedure that supports studies of organ structure and function in biology. During dissection, researchers expose the abdominal cavity, identify the pancreas in relation to nearby organs, and carefully remove or sample its tissue while minimizing damage and contamination. The isolated tissue can then be examined macroscopically, processed for histology, or used in molecular and cellular analyses. This technique helps investigators study pancreatic anatomy, digestive and endocrine functions, disease-related changes, and experimental responses in a well-established mammalian model.

The three-dimensional culture method described in this protocol recapitulates pancreas development from dispersed embryonic mouse pancreas progenitors, including their substantial expansion, differentiation and morphogenesis into a branched organ. This method is amenable to imaging, functional interference and manipulation of the niche.

Mouse Model for Pancreas Transplantation Using a Modified Cuff Technique

Among abdominal solid organ transplantation, pancreatic grafts are prone to develop severe ischemia reperfusion injury-associated graft damage, leading eventually to early graft loss. This protocol describes a model of murine pancreas transplantation using a non-suture cuff technique, ideally suited for analyzing these early, deleterious damages.

We describe a protocol herein for isolating intact islets from neonatal mice. Pancreata were partially digested with collagenase, followed by washing and hand picking. 20 - 80 islet clusters can be obtained per pancreas from newly born mice, which are suitable for several islet studies.

Primary Acinar Cell Isolation: A Method to Isolate Acinar Cells from Mouse Pancreas

This video describes the technique used for isolating pancreatic acinar cells--exocrine cells--containing a variety of digestive enzymes. Primary acinar cells are useful in the ex vivo study of acinar-to-ductal metaplasia (ADM)--an early event in developing pancreatic cancer.
