Neonate Culturing

Neonate culturing is the controlled maintenance of newborn organisms, tissues, or cells under laboratory conditions to support survival, growth, and developmental study. The process relies on aseptic handling and carefully regulated temperature, nutrients, gas exchange, hydration, and environmental cues that preserve viability while limiting contamination and physiological stress. In biology, cultured neonates and neonatal specimens help researchers examine early development, tissue maturation, developmental disorders, and responses to experimental treatments. These models can reveal how genetic and environmental factors shape postnatal biology, while providing controlled systems for investigating mechanisms that are difficult to observe directly in living organisms.

Primary mouse cardiomyocyte cultures are one of the pivotal tools for the investigation of myofibrillar organization and function. The following protocol describes the isolation and culture of primary cardiomyocytes from neonatal mouse hearts. The resulting cardiomyocyte cultures may be subsequently used for a variety of biomechanical, biochemical and cell-biological assays.

We outline methods for the efficient and quick isolation/culture of viable microglia from the neonatal cerebral cortex and adult spinal cord. The dissection and plating of cortical microglia can be accomplished within 90 minutes, with the subsequent microglial harvest taking place ~ 10 days following the initial dissection.

Primary dissociated midbrain dopamine cell cultures allow for the study of presynaptic characteristics of dopamine neurons. They can be used to monitor real-time dopamine release kinetics and protein/mRNA levels of regulators of dopamine exocytosis. Here, we show you how to generate these cultures from rodent neonates.

Epidermal keratinocytes form a functional skin barrier and are positioned at the front line of host defense against external environmental insults. Here we describe methods for isolation and primary culture of epidermal keratinocytes from neonatal and adult mouse skin, and induction of terminal differentiation and UVB-triggered inflammatory response from keratinocytes.

Obtaining a Mixed Glial Cell Culture from a Neonatal Mouse Brain
