Phospholipase C Activation

Phospholipase C activation is a cell-signaling process in which phospholipase C enzymes convert extracellular or intracellular cues into coordinated biochemical responses. In a common pathway, ligand binding activates a G protein-coupled receptor and the Gq protein, which stimulates phospholipase C to hydrolyze the membrane lipid phosphatidylinositol 4,5-bisphosphate (PIP2) into inositol trisphosphate (IP3) and diacylglycerol (DAG). IP3 promotes calcium release from the endoplasmic reticulum, while DAG and calcium activate protein kinase C, altering target proteins and cellular behavior. Studying this pathway helps explain processes including secretion, contraction, metabolism, gene regulation, and signal disruption in disease.

In this video, we demonstrate an assay to establish the presence of phospholipase in the crude venom extract from Anthopleura dowii—a marine sea anemone. The assay utilizes an agarose gel diffusion technique, in which the enzyme phospholipase diffuses and breaks down phospholipids incorporated in the solid media in a radial manner.
