Protamine Sulfate

Protamine sulfate is a positively charged protein-based medication used primarily to reverse the anticoagulant effects of heparin, making it important in cardiovascular medicine, surgery, and biological research. Its mechanism depends on electrostatic binding: the basic amino acids in protamine interact with negatively charged heparin, forming a stable complex that reduces heparin’s ability to enhance antithrombin activity and inhibit clotting factors. Clinicians use this neutralization after procedures such as cardiopulmonary bypass, while researchers study protamine sulfate in coagulation assays, drug delivery, and nucleic acid transfection. Careful dosing is essential because excessive protamine can itself affect hemostasis and trigger adverse reactions.
