Protein Domains

Protein domains are distinct, structurally and functionally specialized regions within proteins that help determine how those proteins fold, interact, and operate. Each domain typically contains a conserved amino acid sequence that folds into a stable three-dimensional structure, enabling activities such as molecular binding, catalysis, or cellular signaling; multiple domains can combine to produce complex functions. In biology, identifying protein domains supports functional annotation, prediction of protein interactions, and analysis of evolutionary relationships. Domain architecture also guides studies of gene mutations and protein engineering, helping researchers investigate disease mechanisms and design molecules with tailored biological properties.

A method for large-scale purification of the APP intracellular domain (AICD) is described. We also describe methodology to induce in vitro AICD aggregation and visualization by atomic force microscopy. The methods described are useful for biochemical/structural characterization of the AICD and the effects of molecular chaperones on its aggregation.
