Protein Transduction

Protein transduction is the delivery of functional proteins into living cells, enabling researchers to alter cellular behavior without introducing DNA. This process commonly uses protein transduction domains or cell-penetrating peptides, which facilitate passage across the plasma membrane and release of the attached protein within the cytoplasm or nucleus. In biology, protein transduction supports studies of signaling, gene regulation, cell differentiation, and disease mechanisms, while also providing a strategy for delivering therapeutic proteins. Because the method can produce temporary effects and avoid genomic integration, it is valuable for experimental manipulation and the development of safer cell-based and regenerative therapies.

We present guidelines for developing synthetic 'chemical transducers' that can induce communication between naturally unrelated proteins. In addition, detailed protocols are presented for synthesizing and testing a specific 'transducer' that enables a growth factor to activate a detoxifying enzyme and consequently, to regulate the cleavage of an anticancer prodrug.
